Transplantation of fetal nigral cells was reported to reverse the 6-OHDA lesion-induced alteration of preproenkephalin, prodynorphin, and preprotachykinin mRNA levels [1,6] and Met-enkephalin levels [19]. From the present study, the nondopaminergic neuromodulation due to intrastriatal transplantation might be related to the reversal of the allodynia-like withdrawal response in 6-OHDA rats.

2012, Progress in Brain ResearchCitation Excerpt :Thus, while amphetamine-induced rotation is usually improved within a couple of weeks of transplantation, suggested to be due to dopamine release from the graft (and dependent on the number of surviving dopaminergic neurons; see Fig. 1), the more protracted improvement in complex motor behaviors is linked to more specific integration of the graft and graft/host interactions. Analysis of intracerebral mRNA expression has shown that some of the parameters of basal ganglia function are improved after transplantation, such as striatal mRNA expression of preproenkephalin, prodynorphin, or D2 receptors (Cenci et al., 1993; Chritin et al., 1992; Savasta et al., 1992; Winkler et al., 2003). Indeed, normalization of the levels of mRNA was also correlated with reduction of LID (Lee et al., 2000), and studies have demonstrated that the number of FosB-positive cells, known to be correlated with severity of LID (Andersson et al., 1999; Winkler et al., 2002), was significantly reduced after dopamine cell transplantation (Carlsson et al., 2009; Lane et al., 2009b; Maries et al., 2003).

Second, the decrease in ppt mRNA expression associated with complete loss of striatal dopamine is reversed by grafts of ventral mesencephalon tissue only in striatal regions that are most densely innervated by the graft (Cenci et al., 1993) – presumably where the graft is capable of generating higher dopamine levels consistent with phasic signaling. Finally, intermittent, but not continuous administration of l-DOPA or a D1 receptor agonist reverses the depletion-induced changes in ppt and ppd mRNA expression, but not ppe (Gerfen et al., 1990; Engber et al., 1991), whereas the opposite holds true for the restoration of ppe mRNA levels by a D2 receptor agonist (Gerfen et al., 1990).
