NMDARs are mechanosensitive (Paoletti and Ascher, 1994) and can be activated by the change of media or plate handling between measurements, which probably led to the increased Ca2+ level observed in the non-stimulated control cells. Hence, Mg2+ was added to the wash-buffer to generate the baseline values.
