Molecular cloning and characterization of a novel phospholemman-like protein from rat hippocampus

Abstract

A cDNA encoding a putative ion channel protein was isolated from a rat hippocampus library. This gene, termed phosphohippolin (Php), contains 318 bp open reading frame encoding a single transmembrane protein with a 5′ signal peptide region. The deduced amino acid sequence shares 48.1% homology with phospholemman (Plm). Expression sequence tag database (dEST) search identified a mouse (AA521976) and human (AA209241) Php gene homologues. The tissue distribution studies of Php mRNA showed its abundant expression in rat brain and kidney, and in the brain, high expression was observed in hippocampus and cerebellum.

    It is especially tempting to speculate that FXYDs were important for the development of a complex nervous system that characterizes vertebrates. FXYD6 (Kadowaki et al., 2004; Yamaguchi et al., 2001) and FXYD7 (Beguin et al., 2002; Crambert et al., 2003), which are prominently and selectively (especially FXYD7) expressed in the brain, probably emerged no later than in the ancestor of Gnathostomata (Pirkmajer et al., 2017), indicating they were important for vertebrate evolution. FXYD1, which is also expressed in the brain (Feschenko et al., 2003), appeared only in tetrapods (Table 4).

    Note that for FXYD1 and FXYD6, interacting αβ-protomers have not been discovered yet (reviewed in Geering, 2008). The differential distributions of Na+/K+-ATPase isoforms and FXYD proteins (Kadowaki et al., 2004; Yamaguchi et al., 2001; Feschenko et al., 2003) suggest distinct roles for these pumps in the respective brain cell types (reviewed in Dobretsov and Stimers, 2005). The functions of the different brain Na+/K+-ATPases (α1Na+/K+-ATPase, α2Na+/K+-ATPase and α3Na+/K+-ATPase) have been extensively studied.
