In contrast, it has been reported that NR1 expression in the TG is decreased in a rat model of masseter inflammation induced by complete Freund’s adjuvant (CFA) intramuscular injections (Lee and Ro 2007). In our study, WB results demonstrated that NR1 protein expression levels were significantly upregulated on days 4 and 7 after IANX, but mRNA levels did not significantly change, possibly due to posttranslational modification (Conradi and Shiu 2018).

It has been demonstrated that NMDAR1 is expressed in both neurons and SGCs in the dorsal root ganglion (DRG)/TG (Castillo et al., 2013; Fernández-Montoya et al., 2017; Lee and Ro, 2007; Schlösser et al., 2015). α2δ-1 is also expressed in the DRG and TG, and trigeminal injury upregulates α2δ-1 protein levels at both locations (Cui et al., 2020; Li et al., 2014; Tachiya et al., 2018).

Lee and Ro87 reported downregulation of the NR1, but not NR2A or NR2B subunits, in trigeminal ganglion neurons after inflammation of the masseter muscle. These findings are consistent with anterograde transport of NMDA receptors to peripheral nerve terminals85,87 after inflammation, suggesting that peripheral NMDA receptors may be involved in inflammatory-induced pain in both the spinal and trigeminal system.
